Guidelines for DNA plasmid preparation and shipping
Drosophila injection
DNA plasmid preparation by Genome Prolab
Success of microinjection is highly dependent of the quality of the plasmid DNA preparation. For this reason, we offer a service of plasmid preparation. If you chose this option, send at least 2 µg of DNA in a minimum volume of 10 µl (200ng/ul) and we will perform the bacterial transformation and the plasmid DNA purification in our facility.
DNA plasmid preparation by yourself
If you chose to prepare your DNA in your laboratory, please read the following guidelines to do so.
For better results, we highly suggest to use Qiagen midi- or maxi-prep kit for your plasmid DNA preparation, since our technology has been optimized with those. We cannot guarantee success of microinjection if you use purification kit from other manufacturer. DNA must be eluted in high-purity water or with QIAGEN elution buffer. However, the dilution of the plasmid should be done with high-purity water and should not be viscous.
VERY IMPORTANT : Microinjection requires at least 25 µg of DNA and we only accept final concentration at 500 ng/µl OR 1000 ng/µl. We do not accept other concentration or plasmids at smaller concentrations.
All samples must be sent in 1.5 ml Eppendorf to facilitate the process.
Molecular Biology services
Targeted mutagenesis
Please send at least 2 µg of plasmid DNA at a concentration of at least 100 ng/µL, in a minimum volume of 20 µL. The DNA must be purified using a column-based plasmid preparation kit.
Guidelines for shipping
Important: Always send your samples with a shipping carrier (Fedex, UPS, Purolator…). Do not ship by regular mail (Canada post).
When sending your plasmid DNA, include the printed submitted form for your samples (step 2 in the Request Our Services section).
Please seal each tube with Parafilm to help prevent leaks. Place the tubes inside a 50 mL Falcon-style tube, then pack it in a padded envelope or box. Dry ice is not necessary for shipping plasmid DNA, but we strongly recommend using an express courier service.
